København 2014.08.19 – Velkommen til en jungle af morskab! De farverige karakterer fra succeskomedien Rio er tilbage med en flok nye venner i denne højtflyvende opfølger for hele familien. Festen fortsætter, da Blu, Jewel og deres tre børn begiver sig ud på en farverig, morsom og musikalsk rejse igennem Amazonas. I sine forsøg på at vænne sig til de nye omgivelser står Blu over for den onde Nigel og den mest skrækindjagende fjende af alle: sin svigerfar!
